I am sending you this document *with regard to* our last discussion (standard).
I am sending you this document *in regard to* our last discussion (standard).
You can use “with regard to” or “in regard to” to indicate the subject that is being talked or written about.

~As regard~ your request, the manager is treating the file (nonstandard).
*As regards* your request, the manager is treating the file (standard).
You can also use “as regards” to indicate the subject that is being talked or written about.
